چکیده

N-dodecyl-N,N-dimethyl-N-benzylammonium halides (DBeAX), like many other amphiphilic quaternary ammonium salts, show biological activity. One of these halides is a major component of "benzalkonium chloride", which is widely used as an antiseptic in pharmaceutical preparations. The mechanism responsible for the antimicrobial action of surfactants is not clearly understood, particularly the role played by counterions in the biological activity of surfactants. In order to elaborate on how counterions affect the properties of biological membranes, we studied the effect of amphiphilic quaternary ammonium salts on the thermotropic properties of the phosphotidylcholine bilayer by means of differential scanning calorimetry. In the case of two series of the halides (N-dodecyl-N,N,N-methylammonium and N-dodecyl-N,N-dimethyl-N-benzylammonium), we found that added surfactants decreased the main phase transition temperature, and that this decrease de pends on the type of counterion. Besides this, we showed that surfactants were able to form surfactant-poor and surfactant-rich domains, and that the kind of counterion influenced domain formation. In this study, the micellization and then interaction of DBeAX (X= Cl-Brand I-) with liposomes formed from DPPC and DPPC containing 5 and 20 mol% of cholestorol were studied by means of high sensitivity isothermal titration calorimetry (ITC). We present and analyse the effect of counterions on the titration curves. The obtained results contribute to a better understanding of the counterion effect in the biological activity of surfactants.

The influence of cholesterol on the interaction between N-dodecyl-N,N-dimethyl-N-benzylammonium halides and phosphatidylcholine bilayers.

Effects of N-dodecyl-N,N-dimethyl-N-benzylammonium halides (DBeAX) on thermotropic phase behavior of phosphatidylcholine/cholesterol bilayers as well as on 1H NMR spectra were studied. The surfactants were added either to the water phase or directly to the lipid phase (a mixed film was formed). The benzyl group, opposite to liposomes without cholesterol, is not incorporated into the bilayer in ...

Counterion effects on interaction of amphiphilic quaternary ammonium salts with model membranes.

The micellization as well as the interaction with model membranes of dodecyltrimethylammonium halides (DTAX) and N-dodecyl-N,N-dimethyl-N-benzylammonium halides (DBeAX) were studied at 298K and 313K by means of titration calorimetry. The calorimetric curves reflect both the counterion and benzyl group effects on the interaction of the surfactants studied with the lipid bilayer. Bromide as count...

Interaction Studies of Sodium N-Dodecyl Sulphate and Protons to Nistone H1

The relationship between the binding of sodium n-dodecyl sulphate and protons to histone H1 has been investigated by equilibrium dialysis and titrimetry. The data cover the pH range 3.2-10 and surfactant concentrations up to 3.0×10-3 mol dm-3. A theoretical approach based on the binding potential concept of Wyman is presented and has been used to make estima...
